The Ribo-Zero™ rRNA Removal Kit (Human/Mouse/Rat) Low Input is ideal for removing rRNA from formalin-fixed paraffin-embedded (FFPE) RNA samples as well as from intact and partially degraded human, mouse, and rat total RNA preparations.

Benefits

  • Removes >98% of 28S, 18S and 5.8S rRNA, and up to 95% of 5S rRNA from intact and partially degraded human, mouse, and rat total RNA preparations including RNA isolated from FFPE samples.
  • Single-pass process yields RNA sufficiently depleted of rRNA for RNA-Seq, poly(A) tailing, random-primed cDNA synthesis, and other applications.
  • Recovers the 5´- and 3´-end fragments of mRNAs, as well as other non-rRNAs, in an intact, partially degraded, or FFPE sample.
  • Uses 100 ng to 1 µg of total RNA.

 

  1. Prepare Ribo-Zero™ Microspheres (15 minutes).
  2. Treat 100 ng to 1 µg total RNA with Ribo-Zero™ rRNA Removal Solution (25 minutes).
  3. Treat sample with and remove Ribo-Zero™ Microspheres (20 minutes).
  4. Purify the rRNA-depleted samples by ethanol precipitation (90 minutes) or column method (provided by the user; 15 minutes).

Figure 1 Figure 2. Agilent Bioanalyzer analysis of Universal Human Reference RNA (UHRR) before and after treatment with the Ribo-Zero rRNA Removal Kit (Human/ Mouse/Rat) Low Input. A 200-ng aliquot of UHRR (Stratagene) was treated with the RiboZero Low Input Kit. A mock-treated sample was prepared from 200 ng of total RNA without the addition of the Ribo-Zero rRNA Removal Solution. After purification, the mock-treated sample was diluted fourfold. Both rRNA-depleted (red) and mock-treated (blue) RNA were analyzed using the Agilent RNA 6000 Pico Kit. Figure 2 Figure 3. Agilent Bioanalyzer analysis of rRNA removal from FFPE RNA.
