Applications

  • Synthesis of RNA for nucleic acid amplification methods or gene expression studies.
  • RNA synthesized can be used as: a hybridization probe, antisense RNA, a ribozyme, a template for in vitro translation, a precursor mRNA for splicing or other processing studies, or to make dsRNA for RNA interference or gene silencing.

T7, T3, and SP6 RNA Polymerases produce defined RNA by in vitro transcription of dsDNA that is downstream of the specific RNA polymerase promoter. These enzyme preparations are highly purified, and exhibit excellent promoter specificity.

Unit Definition: One unit of RNA polymerase catalyzes the incorporation of 1 nmol of ribonucleoside triphosphate into RNA in 1 hour at 37°C under standard assay conditions using a DNA template with the appropriate T7, SP6, or T3 promoter.

Storage Buffer: 50% glycerol containing 50 mM Tris-HCl (pH 7.5), 0.1 M NaCl, 1.0 mM DTT, 0.1 mM EDTA, and 0.1% Triton® X-100.

5X Transcription Buffer (available separately): 0.2 M Tris-HCl (pH 7.5), 50 mM NaCl, 30 mM MgCl2, and 10 mM spermidine.

Quality Control: RNA polymerases are tested in in vitro transcription reactions, and are free of detectable exo- and endonuclease and RNase activities, and E. coli RNA polymerase activity.
